Please have a look at

verify you set the vars correctly.
Then, usually don't work as "root" but alias all root-mail to a "mortal" user,
and work only with that instead of "root" directly (good practice).
Finally, the "inbox" spoolfile must exist to be used of course.
Normally this is created when 1st mail arrives only, not when you start mutt.
Send a mail to "root" first: echo bla | mailx -s begin root
